Putting this in CT because one of the theories clearly is CT. I'll throw that one out first.
1. The CIA, NSA, FBI and all the other usual suspects coming from the "government did it" crowd.
Pretty good case could be made that Hoffa knew too much. And, with Hoffa about to challenge Nixon's imposed lifetime ban barring him from returning to lead the Teamsters upon being pardoned, there is a lot of motive.
Apparently, there was a lot of incriminating evidence found in the Nixon tapes which could serve as ammunition for Hoffa to win a case overturning the ban. Specifically there is on tape a backroom deal between Nixon and Fitz, the Teamsters president at the time, who wanted to keep Hoffa out of the running. And, that would have opened up many more cans of worms, as the video shows.

2. Anthony Giacalone, Detroit Mob "Street Boss"
This is my chief suspect. Not only because he was the guy with the power to order the hit, but also because that's where much of the evidence points.

3. The Irishman, Frank Sheeran.
The only person to confess to the murder. But, if you ask me, he confessed to a lot of things that seem a little over the top.
